摘要
目的:探讨AtheNA定量检测系统在抗核抗体检测中的性能,同时评价其对抗dsDNA抗体检测的敏感性和特异性。方法:收集系统性红斑狼疮(SLE)标本16例,其他自身免疫病标本57例及健康对照标本20例,分别以线性免疫印迹法(LIA法)和AtheNA系统进行抗dsDNA,抗SSA,抗SSB,抗Sm,抗核糖核蛋白(RNP),抗Scl-70,抗Jo-1,抗着丝点B和抗组蛋白9种自身抗体检测;同时以LIA法、间接免疫荧光(IIF)法、ELISA法和AtheNA系统4种方法检测抗dsDNA抗体,评价不同检测方法的性能。结果:LIA法和AtheNA系统对抗核抗体的检测结果显示,健康对照者与SLE患者、不同自身免疫病患者的总体阳性率相比,差异均无统计学意义(P均>0.05)。AtheNA系统与LIA法对9种自身抗体的检测结果具有良好的一致性(P<0.01)。LIA法、IIF法、ELISA法和AtheNA系统4种检测方法对抗dsDNA抗体检测结果显示,AtheNA系统检测抗dsDNA抗体具有较好的敏感性和特异性(68.8%,83.1%),但4种方法对SLE患者和其他自身免疫病患者的检测阳性率差异均无统计学意义(P>0.05)。结论:AtheNA系统对抗核抗体的检测具有良好的性能,其对抗dsDNA抗体的检测具有较好的敏感性和特异性。
Objective : To access the detection property of AtheNA system in antinuclear antibodies and anti-dsDNA antibodies. Methods: Ninety and three serum samples were tested utilizing the AtheNA system and LIA method: 16 samples from patients with systemic lupus erythematosus (SLE), 57 samples from other known autoimmune diseases and 20 healthy donors. The antinuclear autoantibodies to nine different analytes (SSA, SSB, Sin, RNP, Jo-1, Scl-70, dsDNA, Centromere B and Histone) were tested. Meanwhile, the dsDNA antibodies were tested using line immunoassay (LIA), IIF, ELISA and the AtheNA system. Results: There were no significant difference in the antinuclear antibodies detected in three subjects. Antinuclear autoantibodies screening by AtheNA system revealed high concordance with the LIA in samples obtained from 16 samples from patients with SLE, 20 healthy donors, 57 samples from other known autoimmune diseases samples, respectively. The test results of 4 methods showed that the sensitivity and specificity of AtheNA system was better in anti-dsDNA antibody test (68.8% ,83.1% ) than other three methods. Conclusion : The AtheNA system is a sensitive and automatic method for simultaneous quantitative detection of antinuclear antibodies and anti-dsDNA antibodies profiles.
