桂枝汤对糖尿病大鼠右心房自主神经重构的调节作用 被引量:15

Regulation effect of Guizhi Decoction on remolding of the autonomic nervous in the right atrium of diabetes rats

摘要 目的:观察糖尿病大鼠心脏自主神经变化及桂枝汤对心脏自主神经重构的调节作用。方法:55只大鼠,以其中10只作为空白对照组,余45只注射链脲佐菌素(STZ)制作糖尿病模型,4周后,随机分为模型对照组、弥可保组、桂枝汤组,予相应药物灌胃4周。Elisa法检测右心房中酪氨酸羟化酶(TH)、乙酰胆碱酶转移酶(CHAT)、神经生长因子(NGF)、生长相关蛋白(GAP-43)、睫状神经生长因子(CNTF)的含量。结果:模型对照组右心房的TH、TH/CHAT升高、CHAT下降,神经营养因子GAP-43、NGF升高、CNTF降低。与模型对照组相比,桂枝汤组TH下降,CHAT升高,且TH/CHAT降低,神经营养因子CNTF和GAP-43升高,NGF降低,在升高CNTF和抑制NGF方面优于弥可保。结论:糖尿病造成心脏自主神经损伤并重构,桂枝汤能抑制交感神经的异常重构,促进心脏迷走神经恢复,保护自主神经平衡。 Objective: To observe the changes of autonomic nervous in the right atrium of diabetes rats and the regulation effect of Guizhi Decoction on remolding of the autonomic nervous in the right atrium of diabetes rats. Methods: 55 rats in total, with 10 young rats as blank control group, 45 rats as diabetes mellitus (DM) by injection of Streptozotocin (STZ), were randomly divided into model control group, Methycobal group and Guizhi Decoction group, and these rats were administered intragastrically with corresponding drugs for 4 weeks. Enzyme-linked immunosorbent assay (ELISA) was used to detect the levels of TH, CHAT, NGF, CNTF and GAP-43. Results: TH, TH/CHAT, GAP-43 and NGF increased, whlie the CHAT and CNTF decreased in the right atrium of model control group rats. Compared with model control group, Guizhi Decoction elevated the levels of CHAT, GAP-43 and CNTF, while TH, TH/CHAT and NGF decreased. The CNTF increased and inhibition of NGF were better than methycobal group. Conclusion: In the right atrium of DM rats, the cardiac autonomic nervous are damaged and remodeled. Guizhi Decoction can restrain the abnormal remodeling of sympathetic, promote the recovery of cardiac vagal, protect the balance of cardiac autonomic nervous.
