摘要
目的:观察黄芪多糖(APS)对大鼠肾间质纤维化的保护作用,并探讨其作用机制。方法:采用单侧输尿管结扎的方法制造大鼠肾间质纤维化模型,将50只大鼠随机分成假手术组、模型组、黄芪多糖高剂量组(1g/kg)、黄芪多糖低剂量组(0.5g/kg)和氯沙坦组(0.01g/kg),每组10只,给予相应药物治疗21d,假手术组与模型组给予等量0.9%氯化钠溶液。实验末取大鼠左侧肾组织,采用免疫组织化学法检测肾间质中转化生长因子-β1(TGF-β1)、基质金属蛋白酶2(MMP-2)、基质金属蛋白酶抑制因子1(TIMP-1)的表达情况;采用放免法检测组织中血管紧张素Ⅱ(AngⅡ)的表达情况。结果:与假手术组大鼠比较,模型组大鼠TGF-β1、MMP-2、TIMP-1和AngⅡ表达明显升高(P<0.01),黄芪多糖高、低剂量组和氯沙坦组大鼠TGF-β1、TIMP-1和AngⅡ表达明显降低,MMP-2表达明显升高(P<0.05,P<0.01)。结论:黄芪多糖可以在一定程度上抑制肾间质纤维化的形成,保护肾脏,与其上调肾间质中MMP-2表达,下调TGF-β1、TIMP-1和AngⅡ表达有关。
Objective: To investigate the effects of astragalus polysaccharides (APS) on the renal interstitial fibrosis caused by unilateral ureteral obstruction (UUO) in rats. Methods: Rats models of renal interstitial fibrosis were prepared by unilateral ureteral obstruction, and fifty rats were divided into Sham group, Model group, APS high, low dose group and Losartan group. Giving the treatment of corresponding drugs for 21 days, left renal tissue in rats were taken to detect the expression of TGF-β1, MMP-2, TIMP-1 by immunohistochemical method and Ang Ⅱ by radioimmunoassay. Results: Compared with the sham group, the expression of TGF-β1, Ang Ⅱ, MMP-2, TIMP-1 in rats of model group were significantly higher (P〈0.01); Compared with the model group, the expression of TGF-β1, Ang Ⅱ, TIMP-1 in rats of APS high, low group and Losartan group were significantly lower and the expression of MMP-2 were significantly higher (P〈0.05, P〈0.01). Conclusion: APS can inhibit UUO- induced renal interstitial fibrosis in rats, which might be related with the downregulation of the expression of TGF-β1, TIMP-1, Ang Ⅱ and upregulation of the expression of MMP-2.
