习练健身气功八段锦降低戒毒人员CD_4^+ CD_(25)^+调节性T细胞作用的研究

Practicing Qigong Baduanjin on drug addicts can reduce CD_4^+ CD_(25)^+ regulatory T cells

目的:通过比较习练健身气功八段锦前后,戒毒人员外周血中CD4+CD2 5+调节性T细胞(CD4+CD25+Treg)占CD4+T细胞的比例变化,研究健身气功八段锦提高戒毒人员免疫功能的分子机制。方法:80名戒毒人员随机分为健身气功八段锦习练组和对照组各40名。3个月后,流式细胞术法检测习练组和对照组外周血中CD4+CD25+T细胞、CD3+T细胞、CD4+T细胞比例和CD4/CD8比值。免疫磁珠法分离并去除外周血中CD4+CD25+T细胞后,3H-TdR掺入法检测淋巴细胞增殖反应。结果:3个月后,习练组外周血中免疫抑制性CD4+CD25+T细胞比例明显低于对照组(P<0.05);而习练组外周血中CD3+T细胞、CD4+T细胞、CD4/CD8细胞比例明显高于对照组(P<0.05)。习练组外周血T淋巴细胞增殖反应明显高于对照组(P<0.05),免疫磁珠法去除CD4+CD25+T细胞后,对照组外周血T淋巴细胞增殖反应恢复至与习练组和健康对照组水平。结论:习练健身气功八段锦可降低戒毒患者外周血内CD4+CD25+Treg比例,提高戒毒患者的免疫功能。

Objective： To research the effects of Qigong Baduanjin on drug addicts under compulsory addiction treatment （test group）, the CD4＋CD25＋ Treg cells were investigated to discover the mechanism of Qigong Baduanjin to improve the drug addicts＇ immunity. Methods： After practicing Qigong Baduanjin for 3 months, CD3＋T cells, CD4＋T cells, CD4/CD8, and CD4＋CD25＋T cells were tested by flow cytometry. 3H-TdR method was used to test T lymphocyte proliferation response. MACS method was used to delete CD4＋CD25＋ T cells. T lymphocyte proliferation response of CD4＋CD25＋T cells was tested by 3H-TdR method. Results： There were significant difference of CD4＋CD25＋T cells, CD3＋T cells, CD4＋T cells and ratio of CD4/CD8 between test and control after practicing Qigong Baduanjin for 3 months （P〈0.05）. T lymphocyte proliferation response of test group was significant higher than that of heroin addict control （P〈0.05）. While after depleting CD4＋CD25＋T cells by MACS method, T lymphocyte proliferation response of control group was restored similar to test group and healthy control. Conclusion： Practicing JianshenqigongBaduanjin can reduce Treg cells; improve immunity in drug addicts under compulsory addiction treatment.
