摘要
目的观察+Gz暴露条件下大鼠肝脏中热休克蛋白70(HSP70)mRNA和蛋白的变化,探讨+Gz暴露诱导肝脏产生HSP70 mRNA和蛋白的病理生理学意义。方法 100只大鼠随机分为4组,一组为空白对照,其余三组分别进行+2 Gz,+6 Gz和+10 Gz暴露,峰值作用时间3 min,加速度增长率1 G/s。分别于暴露后6 h、12 h、24 h、48 h和96 h麻醉大鼠处死后取肝脏组织,利用RT-PCR和Western blot技术检测HSP70mRNA及蛋白的表达情况。结果与对照组相比,+Gz暴露后6 h、12 h、24 h、48 h,HSP70mRNA及HSP70表达水平升高(P<0.05),暴露后96 h表达水平无差别(P>0.05)。HSP70mRNA及HSP70在+Gz暴露后6 h开始升高,暴露后12 h达到高峰。在同一时间点上,不同+Gz暴露之间比较,+6 Gz组HSP70及HSP70 mRNA表达最高,+2 Gz组次之,+10 Gz最少(P<0.05)。+Gz暴露12h,各+Gz组HSP70及HSP70 mRNA表达水平的变化趋势具有一致性。结论 +Gz暴露增加大鼠肝脏HSP70 mRNA及蛋白表达水平,+Gz暴露环境下机体可能通过代偿机制增加HSP70表达实现对肝脏的保护。
Objective To observe changes of expression level of HSP70 mRNA and protein in rat liver under + Gz exposure, and to explore the physiological and pathological mechanisms of these changes. Methods There were 100 rats divided into four groups randomly. One of the groups was blank control, and the other three groups proceeded in exposure conditions of + 2 Gz, + 6 Gz and + 10 Gz. The peak working time was 3 min, and the growth rate of acceleration speed is 1 G/s. Post + Gz exposure 6 h, 12 h, 24 h, 48 h and 96 h, liver tissues of rats were taken out after anaesthetized and sacrificed immerdiately. The expression levels of HSP70 mRNA and HSP70 in rat livers were tested with RT-PCR and Western blot. Results Compared with the control group, the expression levels of HSP70 mRNA and HSP70 of three + Gz groups were significantly rised at 6 h, 12 h, 24 h, 48 h after exposure of +Gz(P 〈0.05) of HSP70 mRNA and HSP70 had no significant increase (P , but at 96 h after exposure, the expression levels 〉 0.05 ). The expression levels of HSPT0mRNA and HSP70 began to rise from post-exposure 6 h, until 12 h it reached the peak. At the same post-exposure time, the expression levels of HSP70 mRNA and HSP70 under three different + Gz conditions were compared, the +6 Gz group was the highest, the +2 Gz group was the second, the + 10 Gz group was the lowest(P 〈 0.05). Post-exposure 12 h, the expression level of HSP70 mRNA and HSP70 of group +2 Gz, +6 Gz, + 10 Gz had the same trend. Conclusion The expression of HSP70 mRNA and HSP70 in rat liver is increased in post-exposure of + Gz conditions, which can enhance the protection of the liver by a compensatory mechanism to increase the expression level of HSP70.
