摘要
目的探讨环巴胺(Cyclopamine)对人前列腺癌LNCaP细胞增殖和凋亡的影响。方法不同浓度环巴胺(1、5、10、15μmol·L-1)干预LNCaP细胞,分别在不同作用时间(24、48、72h)用MTT法检测环巴胺对LNCaP细胞增殖的抑制,Hoechst染色观察细胞凋亡形态的变化,流式细胞术检测细胞凋亡率的变化。结果环巴胺5、10、15μmol·L-1浓度组对LNCaP细胞增殖有显著抑制作用,与对照组相比差异有统计学意义(P<0.01)。随着浓度的增大对细胞增殖的抑制作用显著增强。10μmol·L-1组于48 h达到半数抑制量(IC50)。环巴胺10μmol·L-1组24、48、72h凋亡率分别为37.21%、57.38%、57.98%,15μmol·L-1组凋亡率分别为21.16%、71.31%、72.90%,与对照组相比差异均有统计学意义(P<0.01)。随着浓度的增大细胞凋亡率逐渐增大。细胞凋亡形态的改变随着环巴胺浓度增大和作用时间的延长而增强。结论环巴胺能够明显抑制LNCaP细胞的增殖,诱导细胞凋亡。
Objective To explore the effect of cyclopamine on proliferation and apoptosis of human prostate cancer LNCaP cell lines in vitro.Methods LNCaP cell lines was treated with cyclopamine in different concentration (1,5,10 and 15μmol · L^-1) and action time of operation (24,48 and 72 hour).The inhibition of cell proliferation was detected with MTT method.The changes of morphology were observed with Hoechst33258 staining method.Flow cytometry (FCM) was used to detect the percentage of cell apoptosis.Results Groups of 5,10,15μmol · L^-1 could inhibit LNCaP cell proliferation,significantly different from that of the blank control group (P <0.01).The group of 10μmol · L^-1 reached IC50 at 48 hour.The apoptosis rates of group 10μmol · L^-1 were 37.21%,57.38%,57.98%,while the rates of 15μmol · L^-1 were 21.16%,71.31%,72.90%,respectively.And there was a significant difference by comparing with the control group(P <0.01).With the concentration of Cyclopamine increased and the role of action time prolong,the cell morphological change of apoptosis was also significantly enhanced.Conclusion Human prostate cancer LNCaP cell lines could be inhibited and be apoptotic by Cyclopamine in vitro.
