CCN1在结肠癌中的表达及其临床意义

Expression and clinical significance of CCN1 in colorectal carcinoma

摘要 目的检测CCN1基因在结肠癌组织中的表达情况,探讨CCN1的表达与结肠癌患者临床病理特征的关系及意义。方法应用Real-time PCR及Western blot方法检测CCN1在4例结肠癌及相应癌旁正常组织中mRNA和蛋白表达水平的差异表达;采用免疫组化检测117例结肠癌石蜡组织中CCN1的表达情况,分析其与结肠癌临床病理特征的关系及预后意义。结果在4例结肠癌组织中CCN1基因在mRNA及蛋白水平的表达高于相应正常癌旁组织;免疫组化检测结果显示CCN1蛋白在结肠癌组织中的表达阳性率为87.2%(102/117),强阳性率为58.1%(68/117),CCN1蛋白在结肠癌组织中的表达水平高于相应癌旁组织;统计学分析结果显示,CCN1蛋白高表达与结肠癌临床分期、淋巴结转移及远处转移密切相关,而与性别、年龄、肿瘤部位、肿瘤大小及分化程度无显著相关性。结论CCN1在结肠癌组织中高表达,其表达水平与肿瘤进展密切相关,提示CCN1在肿瘤发生和转移中可能起到重要作用。 Objective To investigate the correlation between the expression level and clinicopathologic significance of CCN1 in colorectal eancetr,.(CRC). Methods CCN1 mRNA and protein expression in 4 cases of CRC biopsies and their matched adjacent tissues were respectively examined by Real-time PCR and Western blot. Immunohistochemical technique was used to detect the expression of CCN1 protein in 117 cases of paraffin-embedded CRC tissues. Results The concentration of CCN1 was higher in the 4 CRC biopsies than their matched adjacent tissues. Immunohistochemistry and statistical analysis revealed that the positive expression rate of CCN1 protien in paraffinembodded .CRCtissues was 87.2% (102/117) and the Strong positive rate of CCNlprotien was 58.1%(68/117) .The expression of the CCN1 protein was higher in the cancer than in adjacent non-cancerous areas.High CCN1 protein expression was significantly associated with clinical stage, lymph node and distant metastasis, but it was not associated with gender, age, tumor site, tumor size and differentiation. Conclusion Expression of CCN1 is significantly associated with progression of CRC, CCN1 may serve as a valuable molecular marker for prediction of CRC development and metastasis.
