摘要
目的探讨依达拉奉对鱼藤酮诱导的多巴胺细胞的保护作用。方法采用鱼藤酮诱导SH-SY5Y细胞建立帕金森病模型。观察依达拉奉对多巴胺细胞的保护作用;用Hoechst 33258检测细胞凋亡,Western blot检测α-突触核蛋白、B细胞淋巴瘤-2(Bcl-2)蛋白的表达。结果依达拉奉能减少多巴胺细胞凋亡。经过鱼藤酮处理的SH-SY5Y细胞的抗凋亡蛋白Bcl-2降低,而α-突触核蛋白出现过表达及异常聚集;经依达拉奉保护后,Bcl-2的表达上调,而α-突触核蛋白的过表达及异常聚集被抑制。结论依达拉奉能减轻鱼藤酮引起的SH-SY5Y细胞凋亡,并能抑制α-突触核蛋白的表达,上调Bcl-2的表达。
Objective To study the protective effects of edaravone on rotenone-induced dopamine cells. Methods Parkinson's disease SH-SY5Y cell model was established by rotenone and treated by edaravone. The protective effect of edaravone on rotenone-induced dopamine cells was observed by detecting the cell apoptosis with Hoechst 33258 and the expressions of α-synuclein and Bcl-2 with Western blot. Results Edaravone reduced the apoptosis of dopamine cells induced by rotenone. The expression of Bcl-2 was decreaed and α-synuclein was overexpressed and abnormally gathered in SH-SYSY cells induced by rotenone. After treated with edaravone, the expression of Bcl-2 was up-regulated and the overexpression and abnormal gathering of α-synuclein in SH-SY5Y cells were suppressed. Conclusion Edaravone can reduce the apoptosis of SH-SY5Y cells induced by rotenone, inhibit the expression of α-synuclein, and upregulate the expression of Bcl-2.
