青蒿素对糖尿病大鼠肾组织c-jun及c-fos基因表达上调的抑制作用 被引量:2

摘要 目的探讨青蒿素对糖尿病大鼠肾组织c-jun及c-fos基因表达上调的抑制作用。方法 15只大鼠分成正常对照组(A组)、糖尿病非治疗组(B组)及糖尿病青蒿素治疗组(C组),B组、C组应用腹腔单剂量注射链脲佐菌素(STZ)65 mg/kg制备糖尿病大鼠实验模型,C组大鼠给予青蒿素300 mg/(kg·d)腹腔注射。在实验进行到第4周时每组处死大鼠,留取右侧肾脏部分皮质,采用免疫组化及RT-PCR方法分别检测大鼠肾细胞c-jun mRNA、c-fos mRNA及其蛋白表达情况,并观察肾组织细胞的病理改变。结果 B组大鼠肾细胞c-jun mRNA、c-fos mRNA及其蛋白表达显著增加;C组上述指标表达水平分别对2组患者,大鼠肾脏组织病理学改变明显改善。结论青蒿素可通过抑制糖尿病大鼠肾组织细胞c-jun及c-fos基因表达上调而缓解糖尿病大鼠肾脏功能损伤及病理学改变。 Objective It is to approach the inhibitory effect of artemisinin on the upregulation of c -los and c -jun gene expression in kidney tissue of experimental diabetic rats. Methods 15 rats were divide into normal control group (group A) , STZ - induced diabetic group ( group B), and STZ - induced diabetic rats treated with artemisinin ( group C). The diabetic rat model were prepared by using single dose peritoneal injection of streptozocin in group B and group C, then the animals in group C were treated with peritoneal injection of artemisinin. After 4 weeks of treatment, all the animals were sacrificed, and the expressions of c - los and c - jun mRNA and protein levels in kidney of rats were detected by RT - PCR and immunohisto- chemisty staining, respectively. Meanwhile renal pathologic changes were observed. Results The expression of e -los and c - jun mRNA and protein in kidney of rats in group B was significantly elevated after 4 weeks. The abnormal levels of c -los and c -jun mRNA and protein were partly reversed in group C. Accordingly, renal pathologic changes were significantly improved. Conclusion Artemisinin has renoprotective effects in diabetic rats partly through inhibiting the uprcgulation of e -fos and c - jun mRNA and protein in renal cortices.
