摘要
采用细胞病变抑制等方法 ,在植物凝集素 (Phytoagglutinin ,PHA)诱导的草鱼血清中检测到一种干扰素。理化和生物学性质鉴定表明 ,它不同于病毒诱导的α/ β 干扰素 ,表现在对 5 6℃、pH2和 0 .1%SDS敏感 ,其活性不能被α/ β 干扰素抗体所中和 ,其诱生剂为非病毒性质的有丝分裂原 ,诱生作用受佛波酯 (PMA)和白细胞介素 2(IL 2 )的调节 ,这些特性与人类和高等脊椎动物中报道的γ干扰素相符 ,表明是一种γ样性质的干扰素。草鱼γ样干扰素的体内诱导受环境温度、诱导时间、PHA剂量等因素的影响 ,2 5℃或 30℃水温诱导的干扰素活性明显高于 15℃或 8℃。高剂量 (每尾 0 .5~ 0 .8mg)PHA诱导的干扰素活性明显大于低剂量 (每尾 0 .1~ 0 .3mg) ,且达到活性高峰所需要的时间短 ,为 3d ,而低剂量为 5d。PMA和IL
An interferon factor was detected in grass carp (Ctenopharyngodon idellus) serum, induced in vivo with phytoagglutinin by cell pathological effect inhibition technique. Study on its biochemical and biological characteristics showed that it was different from the interferon α/β induced by virus. It was sensitive to heat (56℃), pH2 and 0.1% SDS, and its activity could not be neutralized by the antibody against interferon α/β. It proved that the inducer was mitogen but with non virus character, and the production could be modulated significantly by the phorbol myristate ester and interleukin 2. All these were consistent with the interferon γ in human, which indicated it was interferon γ like activity. The induction was influenced by the temperature, inducing time and dosage of phytoagglutinin. The activity was higher at the temperature of 25℃ or 30℃ and at phytoagglutinin dosage of 0.5-0.8 mg per fish than that at 8℃ or 15℃ and at that of 0.1-0.3 mg per fish. The inducing time when the activity was the highest was 3 days at high dosage of phytoagglutinin, which was shorter than that of 5 days at low dosage. The interferon induction could be promoted by the phorbol myristate ester and interluekin 2.
