摘要
采用单克隆抗体作为试剂,建立了直接用于快速检测粪便中福氏痢疾杆菌的一种新方法即固相致敏红细胞吸附技术(SPASE)。检测210份疑似细菌性痢疾患者粪便标本的结果证明,用常规培养法检测阳性者,SPASE法检测均阳性,两类方法的符合率为81.9%;常规培养法阳性率43.3%而SPASE法则为61.5%(P<0.001)。SPASE法全过程仅需2.5~3小时,可检出103~104个/ml细菌,这种快速免疫法不仅能检出完整的细菌,而且也能检出其裂解产物,从而证实了SPASE法具有较高的敏感性、特异性和快速性,且不需复杂的仪器设备,加之实验程序简单,甚适于医院及基层医疗单位应用。
olid-phase absorptiOn to sensitized erythrocytes(SPASE )test is a new techniqueof solid-phasc aggregation,Using antisensitzed erythroeytes as indicator to replace enzymeor isotoqe,the result is judged by observing the agglutination of erythrocytes,A micro-titre plate is coated by anti-LPS McABs of Shigella flexneri.After heating and wash-ing,the boiled fecal sample is added to the microtitre plate. The McABs sensitized ery-throcytes is put into the microtitre plate lastly.“++” agglutination is considered posi-tive.The results showed that the whole detection process by SPASE required only 2.5~3h,its sensitivity was l03~104 bacteria/ml,it is a specific method for detection ofShigella flexneri and its’very convenint.In addition,it requires only a small amountof rcagent,no spccial apparatus are needed.With all these advantages,we considerthat SPASE is an ideal technique for rapid detection of Shigella flexneri, sonnei fromfecal samples,particularly for rural medical units.
