摘要
本文建立了反相高效液相色谱法测定安定的血药浓度。柱C18,流动相为甲醇-水(71:29),水中含0.14ml正丁胺和0.09ml冰醋酸);检测波长230nm。在0.1~3μg/ml浓度范围内线性良好,最低检测限为50ng/ml。平均回收率为88.19±0.5%。用本法测定了分娩产妇iv注安定后的体内血药浓度,并进行了药代动力学参数计算。
A Rp-HPLC Method has bee developed for the determination of diazepam(DZP) in serum.Chromatographic separation has been achieved on C18 column with methanol-water (71:29, water contains 0.14ml n-butylamine and 0.09ml acetic acid glacial) as the mobile phase. Detection was set at UV 230nm. The lower limit of detection of DZP in serum was 50ng/ml. The calibration curve was linear between 0.1 and 3μg/ml with a regssion coefficient of 0.9988. This assay procedure has been applied to monitoring serum levels of diazepum in parturients who received IV administration of drug.The pharmacokinetics of diazepum could be described by a two compartment open model.Its parameters were as follows:t1/2=4.87±2.09min,t1/2=184.72±36.59min,AUC=151.05±42.20μg/ml/min.
