摘要
目的探讨银杏叶提取物( Egb761)对 SD乳鼠心肌细胞外钙内流的影响。方法选择三天鼠龄的雄性 SD乳鼠心肌,按 Hoffman法行心肌细胞培养 10 12天。实验分二组:对照组和实验组( Egb761分成三种浓度 10- 3mol、 10- 5mol、 10- 7mol)。应用 Fura 2荧光探针的方法,观察各组静息状态和加氯化钾(浓度为 20mmol)后的心肌细胞游离钙浓度的改变。结果( 1)基础状态下,氯化钾能促进心肌细胞外钙内流,使心肌细胞内游离钙浓度增高 (P<0.001);( 2) Egb761对氯化钾激发的细胞外钙内流的增加有抑制作用 (P<0.05)。结论 Egb761具有抑制 SD乳鼠心肌细胞外钙内流的作用,提示 Egb761可能对钙超载所致的心肌细胞损伤具有防治作用。
Objective To detect the effect of Gingko biloba extracts(Egb761) on the inflow of extracellular calcium ions in SD new born rats myocardium.Methods The myocardium of three day old male SD rats was used in myocardiocyte culture.The experiment was divided into two groups:the placebo group and the test group in which the Egb761 was given by three concentrations 10- 3mol、 10- 5mol、 10- 7mol.In each group the concentration of intramyocardiocellular separating calcium ions were measured by Fura 2 probe both at the base state and after adding KCI.Results (1)KCI could elavate the inflow of extramyocardiocellular calcium ions at the base situation,which would increase the concentration of intramyocellular separating calcium(P<0.001).(2)The Egb761 could inhibit the increase of inflow of extramyocellular calcium ions activated by KCI(P<0.05).Conclusions It was suggested that Egb761 could prevent myocardium from injuring caused by the overloading of calcium ions.
