摘要 建立了反相高效液相色谱法测定血清中氨甲蝶呤浓度的方法。以 C1 8为固定相 ,乙腈 -醋酸缓冲液 (0 .1 m,p H4.0 ) -水 (3∶ 1 0∶ 7)为流动相 ,检测波长 2 94nm,以咖啡因为内标 ,经乙酸乙醋提取浓缩后 ,MTX在 0 .0 5~ 2 5μg· ml- A RP-HPLC method is described for the determination of MTX in plasma on C 18 column using acetonitrile-acetate buffer(0 1m,pH4 0)-water(3∶10∶7)as mobile phase. The UV detection wavelength was 294nm. MTX was extracted by ethyl acetate used coffaine as internal standard and the linearship appeared excellently within the range of 0 05~25μg·ml -1
