维甲酸抑制人胃癌细胞株层粘素受体基因和原癌基因C-Ha-ras mRNA的表达

摘要 目的:研究维甲酸(RA)对人胃癌细胞株(SGC7901)层粘素受体(LNR)基因和原癌基因C-Ha-ras表达的影响。方法:应用原位杂交方法标记LNR探针和C-Ha-ras探针,对人胃癌细胞(分为经RA处理及未经RA处理两组)中LNR和C-Ha-ras基因的mRNA表达进行检测。结果:经RA处理的人胃癌细胞LNR mRNA表达较未经RA处理者显著降低(P<0.01~0.001);C-Ha-ras转录水平亦显著降低(P<0.01~0.001)。结论:RA可显著抑制人胃癌细胞LNR和C-Ha-ras基因的表达。 Background/Aims: To investigate the expression of laminin receptor (LNR) gene and protooncogene C-Ha-ras in human gastric cancer cell line (SGC7901) induced differentiation with retinoic acid (RA). Methods: The cell hybridization was used to detect the mRNA expression of LNR and C- Haras gene of SGC7901 treated/untreated with RA. Results: The mRNA expression levels of LNR and CHa-ras were significantly decreased in the cells treated with RA than those untreated ones (P<0.01- 0.001). Conclusions: RA could highly inhibit the expression of both LNR and C-Ha-ras gene in human gastric cancer cells.
