摘要
目的 :观察神经节苷脂对损伤脊髓神经组织的保护作用 ,并初步探讨其作用机制。方法 :重度脊髓损伤大鼠随机分对照组及神经节苷脂治疗组 (n =15 )。在蛛网膜下腔注射生理盐水 2 0 μl或神经节苷脂 30 μg。观察体重变化、局部血流量、凋亡细胞原位标记、神经功能评价和定量组织学分析。结果 :局部血流量 2组无明显差异 ,治疗组存活率、体重、神经功能评分和残余神经组织面积明显高于对照组 ,而TUNEL阳性细胞数量明显低于对照组。结论 :神经节苷脂对损伤脊髓组织有明显保护作用 ,可能是通过对神经细胞凋亡的阻断来发挥作用的。
Objective:To study the protective effects and the mechanism of monosialicganglioside (GM) on the spinal cord injury.Methods:Following a controlled,severe degree compression injury to the lower thoracic spinal cord(T 8,9 ),rats were divided into tow groups randomly.One group of rats were treated with normal saline solution(20μl) intrathecally 15 minutes after injury.The other rats were administrated GM(30μg,20μl) intrathecally.The changes of weight,spinal cord blood flow(SCBF),the terminal deoxynucleotidy1 transferase-mediated dUTP nick end labeling (TUNEL),neurological function and quantitative histological sparing were observed.Results:We found that GM had little effects on the SCBF.But GM could improve the neurological function,weight,area of spared normal tissure,and obvioulsy reduce the mortality and the number of TUNEL-positive cells.Conclusion:The results suggest that GM has neuroprotective effects on the injured spinal cord,the mechanism of the beneficial effects of GM may be resulted from reduction of spinal cord cell apoptosis.
