老年期肾阴阳虚损大鼠的线粒体能量代谢及其调控机制的研究 被引量:11

摘要 目的 :观察老年期肾阴阳虚损大鼠的线粒体能量代谢变化及其调控机制。方法 :将 2 4月龄大鼠按掌温高低区分为老年“肾阴虚”(掌温高 )、老年对照 (常温 )和老年“肾阳虚”(掌温低 )三组 ,采用氧电极法和放免法等技术观察各组老年鼠肝线粒体呼吸功能、氧化酶活性、游离钙、MDA浓度、肝Na+-K+-ATP酶活性和血清FT3、FT4 、T、下丘脑NE含量以及心率等变化。结果 :老年“肾阳虚”鼠RCR、P O、OPR、细胞色素C氧化酶、Na+-K+-ATP酶活性、线粒体游离钙和血清T、FT3、FT4 及下丘脑NE含量均低于“肾阴虚”鼠 ,而“肾阳虚”组MDA含量却明显升高。结论 :老年“肾阳虚”鼠线粒体能量代谢低下 ,氧化损伤加剧 。 Objective:To research on the energy metabolism of mitochondris and its regulation of aging rats with “deficiency of Kidney Yin or Kidney Yang”. Methods:According to the post pedes temperature, the SD senile rats were divided into “kidney yang deficiency” group、control、“kidney yin deficiency” group. The liver mitochondrial respiratory function the activity of oxidase MDA and free calcium concentration liver Na +-K +-ATPase, serum free triiodinethyroxine(FT 3)、free tetra iodinethyroxine(FT 4)、testosterone(T)、hypothalamus NE content and heart rate、were estimated by the oxygen electrode method and radioimmunoassay etc. Results:The liver mitochondrial respiratory function、the activity of oxidase、free calcium concentration、liver Na +-K +-ATPase, serum free triiodinethyroxine(FT 3)、free tetra iodinethyroxine(FT 4)、testosterone(T)and hypothalamus NE content in “kidney yang deficiency” group were significantly decreased than the “kidney yin deficiency” group, mitochondrial MDA content of “kidney yang deficiency”, however, was higher obviously. Conclusion:The energy production and energy expenditure of “kidney yang deficiency” group were lower, oxidative damage were more serious than “kidney yin deficiency” group and was correlated to the mitochondrial self regulation and neuro endocrinal regulation.
