摘要
目的 研究视黄酸 (RA)对胃腺癌MGC - 80 3细胞的作用及有关机制。方法 以荧光显微镜、透射电镜和电泳技术检测细胞凋亡 ,免疫组化检测细胞P 2 1/WAF 1蛋白表达。结果 RA处理的MGC - 80 3细胞发生凋亡特征性的改变 :细胞皱缩 ,核染色质凝集 ,边移 ,沿核膜内缘分布 ,呈帽形或半月形 ,亦可见凋亡小体的产生 ;基因组DNA沿核小体之间断裂 ,电泳得到凋亡特征性的DNA梯带 ;细胞P 2 1/WAF 1蛋白表达显著增高。结论 RA诱导胃腺癌MGC - 80 3细胞凋亡与其上调P 2 1/WAF
Objective To investigate whether and how retinoic acid(RA) can induce apoptosis of MGC-803 gastric adenocarcinoma cells.Methods Morphological changes of apoptosis were observed under light microscope and transmission electron microscope.DNA fragmentation was assessed by agarose gel electrophoresis.The level of P 21/WAF 1 protein was measured by immunocytochemistry.Results After 3-5 days of exposure to all-trans RA,a ladder appeared pattern characteristic of apoptosis in MGC-803 cells.Moreover,RA increased the P 21/WAF 1 gene expression.Conclusions RA can induce apoptosis of MGC-803 cells mainly via upregulation of P 21/WAF 1 gene expression.
