摘要
目的 :CDKN2 (P 16 /MTS 1)是细胞周期抑制基因和抑癌基因。探讨该基因产物P 16 /MTS 1在大肠癌及其对照组织中的表达与大肠癌发病的关系及对预后的预测价值。方法 :应用S P免疫组织化学染色技术和图像分析技术对 2 5例大肠癌、10例大肠腺瘤、8例正常大肠石蜡包埋组织切片进行P 16 /MTS 1定性和定量分析 ,并观察P 16表达与大肠癌发病及术后 5年生存率的关系。结果 :P 16在大肠癌、大肠腺瘤、正常大肠组织表达阳性率分别为 6 4%、6 0 %、2 5 % ,染色灰度分别为132 .16 1± 1.5 42、147.415± 2 .5 48、15 1.910± 2 .86 3(P <0 .0 5 ) ,P 16表达阳性和阴性的大肠癌患者术后 5年生存率分别为81.2 5 %和 6 6 .6 7%。结论 :提示P 16在大肠癌的表达与大肠癌的发病相关 ,可能成为大肠癌预后有价值的指标之一。
Aim: CDKN 2 is a cell cycle inhibitor and tumor suppressor gene. The purpose of this study is to investigate the association between expression of CDKN 2 gene products(P 16/MTS 1) and pathogenesis of colorectal carcinoma to predict its value in the prognosis of tumor. Methods: To apply S P immunohistochemistry staining technique; to assess paraffin embedded tissues sections of 25 cases of colorectal carcinomas, 10 cases of adenomas and 8 cases of normal colorectal with CDKN 2 (P 16/MTS 1) qualitative and quantitative analysis; to observe the relationship between the expression of P16 and colorectal tumor pathogenesis and 5 year survival rate post operation. Results: Positive expressive rate of P16 was 64% (16 of the 25 cases) for colorectal carcinomas 60% (6 of the 10 cases) for colorectal adenomas and 25% (2 of the 8 cases) for normal colorectal tissues. Computerized photography counted analysis were 132.161±1.542, 147.415±2.548, 151.910±2.863 respectively (P<0.05). A 5 year survival rate of the patients with P16 positive and negative expression were 81.25% and 66.67% respectively. Conclusions: These results indicate that P16 expression might be associated with occurrence of colorectal cancer and might be a valuable index for the prognosis of colorectal carcinoma.
