碱性成纤维细胞生长因子对大鼠局灶性脑缺血再灌注损伤神经细胞凋亡的调控 被引量:5

摘要 目的探讨碱性成纤维细胞生长因子 (bFGF)在局灶性脑缺血再灌注损伤中对缺血后神经细胞凋亡的调控机制及作用。方法在缺血及再灌注不同时间点,采用免疫组化及逆转录-聚合酶链反应 (RT- PCR)方法检测缺血中心区及半影区内 bFGF蛋白及 mRNA表达的动态变化。结果缺血半影区 bFGF蛋白及 mRNA达表明显增加,缺血中心区表达略有增加。结论 bFGF的表达趋势与缺血再灌注损伤时神经细胞凋亡趋势相符,说明 bFGF对神经细胞有保护和修复作用。 Objective To observe the effect and regulating mechanism of bFGF on nerve cell apoptosis during reperfusion after focal cerebral ischemia.Mehod The dynamic expression of bFGF and mRNA was examined during ischemic and reperfusion period by reverse- transcript PCR and immunohistochemical technology in sichemic core and penumbia. Method Using a focal cerebral ischemic model with reperfusion and immunogisto- chemistry method,RT- PCR techonology.Result The significant expression of bFGF and mRNA were observed in penumbia,while only slight increase in expression of bFGF and mRNA was observed in ischemic core.Conclusion Expression of bFGF is in accordance with nerve cell apoptosis during reperfusion after cerebral ischemia indicating bFGF played and important role in protecting and promoting recovery of nerve cells.
