摘要
目的检测与支气管哮喘遗传相关的基因。方法将门诊或病房确诊为哮喘的病人按实验要求分为两组,一组为有明显家族遗传史的患者,另一组为没有家族史的散发患者。首先从实验对象的外周血中提取 DNA, PCR扩增 IL- 4的第二个内含子中的一段 TG重复序列,然后用 8%的聚丙烯酰胺凝胶电泳检测这段基因的多态性。结果在这段基因序列中检测出 4种多态性,其中等位基因表型 1和等位基因表型 2在有遗传史的哮喘患者与无遗传史的哮喘患者中出现的频率有区别。结论 IL- 4的第二个内含子中的 TG重复序列的多态性可能与哮喘的遗传有关。
Objective to detect polymorphism of asthma related genes. Methods Patients diagnosed asthma were divided into two groups ,one group has genetic susceptibility ,the other has no such character.DNA were extracted from the peripheral blood,then TG sequence in second intron of IL- 4 was amplified by PCR, the polymorphism of that segment were then detected through 8% polyacrylamide gel.Result four kinds of penotype were found in such segment.The frequency of phenotype 1 and type 2 of in test group are statistically different from that in the check group. But the other two phenotype frequencies are just the same in two groups.Conclusion the different frequency of phenotype 1 and phenotype 2 in two groups may give us some meaning in asthma genetics.
