摘要
目的 研究血管紧张素转换酶 (ACE)基因多态性与胰岛素抵抗 (IR)及血脂之间的相关关系。方法 应用PCR技术和遗传学方法 ,检测正常人 66例和冠心病、高血压和 2型糖尿病 176例患者的ACE基因多态性 ,测血糖、血脂、胰岛素等指标 ,计算ISI,分析ACE基因多态性与IR及血脂之间的关系。结果 疾病组和对照组在性别、年龄构成方面差异均无显著性 (P >0 .0 5)。①血压、CH ,TG ,LDL c水平明显高于对照组 (P <0 .0 5) ,而HDL c水平明显低于对照组 (P <0 .0 5) ,且ACE基因DD型组与ID ,II型组相比差异有显著性。②疾病组IR的发生率为 2 7.6% ,其中DD基因型占 57.1% .IR组的DD基因型频率和D等位基因频率显著高于II基因型频率和I等位基因频率 (P <0 .0 1,P <0 .0 1)。结论 ACE基因DD型和D等位基因与IR及血脂代谢异常有关 。
Objective To study the relationship of the angiotersin converting enzyme(ACE)gene polymorphism and insulin resistance(IR)and plasm lipid.Method 176 patients with coronary heart diseases or hypertensive or 2 diabetes mellitus and 66 unrelated healthy individual were studied by using polymorase chain reaction(PCR)and genetic methods.Blood glucose or plasm lipid and insulin were performed and insulin sensitivity index(ISI)was calculated.To explore the relation ACE gene polymorphism and IR and plasm lipid.Result There was no significant difference in sex and age between patients group and normal control group(P>0.05).①Blood pressure ,CH ,TG and LDL c level of the patients group was higher than that in the normal control group(P<0.05),but HDL c level was lower(P<0.05).The CH ,TG,LDL c and HDL c level of ACE gene DD genotype group was significant difference than those of ID and II genotype group.②The total prevalence of IR in patients group was 27.6%,The frequencies of the DD genotype was 57.1%.The frequencies of the DD genotype and D allele in IR group were significantly higher than those of the II genotype and I allele(P<0.01,P<0.01).Conclusion DD genotype and D allele is closely related to IR and abnormal plasm lipid metabolism,but I allele is not.
