摘要
目的 :探讨纯化外周血T淋巴细胞的 3种方法及其特点。方法 :分别应用尼龙毛法、补体细胞毒法和免疫磁珠法分离健康人外周血。结果 :尼龙毛法分离后T细胞纯度为 ( 70 7± 2 8) % ,明显高于分离前的 ( 5 5 6± 3 7) % (P <0 0 5 ) ;补体细胞毒法为 ( 82 8± 3 2 ) % ,显著高于尼龙毛法 (P <0 0 5 ) ;免疫磁珠法为 ( 96 0± 1 4) % ,最高纯度可达 98 1% ,显著高于尼龙毛法和补体细胞毒法 (P <0 0 5 )。免疫磁珠法分离后T细胞的回收率为 ( 91 6± 6 8) % ,显著高于其它两种方法。结论 :尼龙毛法和补体细胞毒法能够有效地初步分选T细胞。免疫磁珠法是高效纯化T细胞的新方法。
Objective:To evaluate the effects and characteristics of three methods on purifying T lymphocytes.Methods:Human peripheral T lymphocytes were separated by nylonwool,complement cytotoxicity and magnetic activated cell sorter(MACS) respectively.Results:The purity of T cell by three methods was (70 7±2 8)%,(82 8±3 2)% and(96 0±1 4)% respectively. These purities were all significantly higher than those before separating. MACS can purify T cell to 98 1% and it was significantly higher than other two methods(P<0 05).Conclusion:Nylonwool and complement cytotoxicity can purify T cell effectively and primarily. MACS is a new purifying method of high efficiency.
