摘要
目的 :探讨细胞凋亡和P5 3蛋白在宫颈上皮内瘤样病变 (CIN)和鳞癌中的表达。方法 :采用DNA末端缺口标记技术和免疫组织化学技术检测 15例正常宫颈组织、14例CINⅠ、10例CINⅡ、2 0例CINⅢ和 5 2例浸润性宫颈鳞癌 (ISCC)中细胞凋亡标记指数 (A LI)和增殖细胞核抗原标记指数 (PCNA LI)。结果 :在正常宫颈组织未检测到凋亡细胞 ,从CINⅠ→CINⅡ→CINⅢ ,A LI逐渐增加 (P <0 0 5 ) ,ISCC中A LI明显低于CIN(P <0 0 1) ,PCNA LI随宫颈癌各级病变的进展逐渐增加 (P <0 0 5 ) ;在正常宫颈和CINⅠ ,未检测到P5 3蛋白表达 ,在CINⅡ可检测到少量阳性细胞 ,P5 3 LI在CINⅢ明显低于ISCC(P <0 0 1)。结论 :细胞凋亡在宫颈癌发生发展中发挥了重要作用 ,P5 3蛋白的调节可能阻断了细胞凋亡。
Aim:To probe the expressions of apoptosis and P53 gene in cervical intraepithelia neoplasia and squamous cell carcinomaMethods: The apoptotic labeling indices(A-LI),PCNA(proliferating cell nuclear antigen)-LI,P53-LI and the positive expression of P53 protein were observed in 15 cases of normal cervical epithelia, 14 cases of CINⅠ, 10 cases of CINⅡ,20 cases of CINⅢ and 52 cases of ISCC (invasive squamous cervical carcinoma) tissues, using DNA nick end labeling technique and immunohistochemical staining for PCNA and P53 proteins Results:No apoptotic cell was observed in normal cervical epithelium, A-LI increased with CINⅠ→ CINⅡ→CINⅢ(P<005), A-LI in ISCC were significantly lower than those in CIN(P<001) PCNA-LI gradually increased with the development of every lesion grade of cervical carcinomas (P<005)P53 protein was not detected in normal cervical epithelia and CIN I, but a few in CINⅡ, and P53-LI in CINⅢ were significantly lower than those in ISCC(P<001) Conclusion: During the development of cervical carcinoma ,apoptosis may play an important role, and the regulation of P53 protein might block apoptosis
