摘要
目的 研究中枢α2 去甲肾上腺素受体在大鼠点燃模型的作用。方法 建立大鼠杏仁核点燃和戊四唑 (pentylenetetrazol)点燃模型 ,测定α2 受体激动剂可乐定 (clonidine) 0 .0 1~ 5 .0mg·kg-1ip对发作行为和后放电的影响 ,并用α2 受体阻断剂育亨宾(yohimbine) 2mg·kg-1ip预处理 ,再观察可乐定对点燃的影响。结果 可乐定对大鼠杏仁核点燃和戊四唑点燃均呈剂量依赖性抑制作用 (P <0 .0 1) ,育亨宾完全反转可乐定对点燃的抑制作用。结论 中枢突触后α2
OBJECTIVE To study the role of central α 2 noradrenoceptors in kindling models in rats.METHODS It was established the amygdala kindling model and pentylenetetrazol (PTZ) kindling model in rats.The agonist of α 2 noradrenoceptors,clonidine 0.05~5.0 mg·kg -1 was given by ip.The inhibitory effects on the behavior and afterdischarge of kindled rats was observed.After pretreated with antagonist of α 2 noradrenoceptors yohimbine 2 mg·kg -1 ip,the effects of clonidine on kindled rats was then observed.RESULTS Clonidine could dose dependently inhibit the seizure severity in both kindling models( P <0.01).The inhibitory effect could be reversed completely by the pretreatment with yohimbine.CONCLUSION Central postsynaptic α 2 noradrenoceptors may play an important role in amygdaloid kindling and PTZ kindling models in rats.
