应用全自动角膜内皮细胞分析仪对保存兔角膜内皮细胞定量研究 被引量:1

摘要 目的 :研究保存的兔角膜内皮细胞活性四项定量指标的变化。方法 :应用非接触式眼库镜及计算机角膜内皮细胞形态定量分析系统对 42只 (84片 )实验兔角膜内皮细胞进行四项指标 (平均细胞面积、细胞密度、六棱细胞比率、变异系数 )定量分析 ,并同时检测内皮细胞活性率 ,其结果进行对比研究。结果 :改良 K3液组与 K液对照组 :保存 5天时 ,四项指标两组无显著性差异(P>0 .0 5 ) ,保存 7、 10天时 ,四项指标两组有显著性差异 (P<0 .0 5 )。改良 K3液组及 K液组分别与正常兔组进行比较 :改良 K液组在保存 10天时 ,四项指标与正常兔组差异有显著性 (P<0 .0 5 )。K液对照组在保存 7、 10天时 ,四项指标与正常兔组差异有显著性 (P<0 .0 5 )。改良 K3液组在保存 10天时 ,内皮细胞活性率在 93.36± 1.96 % ,而四项指标与正常兔组差异已有显著性。K液对照组在保存 7、 10天时 ,内皮细胞活性率分别为 90 .84± 1.9% ,79.8± 4.48% ,四项指标均与正常兔组差异有显著性。结论 :完整的评价角膜植片的活性 ,单纯依靠既往的内皮细胞活性率测定是不全面的 ,在有条件的情况下应同时结合四项定量指标进行综合评价 ,才能确保供体角膜片的质量。 To investigate the quantitative changes in the four indexes of the viability of preserved rabbite corneal endothelium.Methods:Apply the non-contact donor lens and the computeraided endothelial quantitative morphological analysis system to the rabbit corneal endothelium(n=84)for quantitative analysis of four indexes(average cell area,endothelial cell density,coefficient of variation of cell area,ratio of hexagonal cells),as well as the endothelial cell viability rate measurement of the endothelium,the results is used for comparison.Results:Improved K 3—sol group and K—sol control group:for 5 days,there were no significant difference(P>0.05)in the four indexes,for 7、10 days,there were significant difference(P<0.05)in the four indexes.Improved K 3—sol group、K—sol control group and normal rabbit group:①Improved K 3—sol group and normal rabbit group,for 10 days,there were significant difference(P<0.05)in the four indexes.②K—sol control group and normal rabbit group,for 7、10 days,there were significant difference(P<0.05)in the four indexes.Improved K 3—sol group at 10 days,the endothelial cell viability was 93.36±1.96%,but the four indexes were significantly different between K 3—sol group and normal rabbit group.K—sol control group at 7、10 days,the endothelial cell viability were 90.84±1.9%、79.8±4.48%,the four indexes were significantly different between K—sol control group and normal rabbit group.Conclusions:Without the endothelial cell viability rate measurement,the evaluation of the corneal graft viability is relatively not perfect,the four quantitative indexes should be included for considerateness.in this way,the quality of donor graft can be better ensured.
