摘要 布洛芬[2-(4异丁基苯基)丙酸]为临床上广泛使用的抗炎、抗风湿药。目前国内正在研制该药的控释剂型,以期克服普通片,药效短、服用剂量大等缺点。建立血中布洛芬的分析方法目的是评价各种不同剂型在人体内的差异,籍此作为选择合适的控释剂型的依据。同时,该法亦可用于临床的血药浓度监测。 A method for the determination of ibuprofen in plasma was developed. Ibuprofen in 0,5ml of heparinized plasma which was acidified with 0.2ml of 4mol/L HCI was extracted with 5ml of hexane-isopropyl alcohol (9:1). The organic phase was dried and redissolved in 1.0ml of mobile phase and 20μl of which was injected into a column of Nucleosil C3 (φ4.6×200mm, 5μ), then eluted with mobile phase metbaaol-water-0,049 mol sodium phosphate buffer (220:100:10v/v)and were detected under 2l4nm. The linear range of the method was 0.2-40μg/ml and the coefficients of variation of the intra- and inter-day assay were within 3% and 7% respectively. The method is simple and accurate, and has been successfully used for biopharmaceutical investigation of ibuprofen.
