摘要
本试验采用阳离子化牛血清白蛋白(C-BSA)制作大鼠膜性肾病模型。大鼠随机分四组,除正常对照外,其余三组在往射C-BSA的同时,分别注射等渗盐水、去铁敏(DFO)和人参皂甙,共5周。结果DFO和人参皂甙能显著降低蛋白尿,降低血清及肾皮质中脂质过氧化终产物丙二醛(MDA)含量,提高血清及肾皮质中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-px)活性,组织学检查显示DFO和人参皂甙轻度改善IgG,C3和BSA沉积,显著改善上皮细胞肿胀和足突融合。提示中药提取物人参皂甙与羟自由基清除剂DFO相似,能抗脂质过氧化,保护抗氧化酶活性,降低C-BSA大鼠蛋白尿。
he purpose of this experiment was tostudy the role of ROM in
C-BSA-inducedMN of rats and the protective effect of Gin-senoside(GS) . 31 Wistar rats were
random-ly assigned to 4 groups ,each preimmunizedsubcutaneously with mg C-BSA
emulsifiedin incomplete Freund's adjuvant. Seven dayslater, eontrol group received saline
only,other groups received intraveneous C-BSA3. 5 mg every other day until the end of the4th
week. Saline, DFO and GS were givenseperately in different groups before the in-jection of
C-BSA and continued during anti-gen challenge. DFO and GS treatment re-sulted in significant
decrement of protein-nuria compared with control. DFO and GSalso resulted in significant
decrement ofMDA content (P<0. 01 ) and increment ofSOD and GSH-px activities (P < 0. 01
inserum and renal cortex) as compared withthe control. If study showed that DFO andGS
slightly attenuated CBSA rats glomeru-lar IgG,C3 or BSA deposition. LM showedthickening and
spikes of GBM in experimen-tal model but a mild extent in DFO and GS-treated rats. EM
showed DFO and GS coulddecrease subepithelial electron-dense de-posits and attenuate
fusion of foot process.Our results suggested that ROM might me-diate in part glomerular injury
in C-BSA-in-duced MN. GS could reduce the extent oflipid peroxidative injury , protect the
activityof lipid peroxidative injury, protect the ac-tivity of anti-oxidase and recuce proteinuri-a .
