Effect factors of measurement to cholera antitoxin with ELISA-inhibition technique were discussed. The optimal coated concentration of cholera antitoxin IgG was 5μg/ml, incubated temperature was 12 h at 4 C or 1 h at 37℃. One per cent bovine serum albumin and 0.5% Tween 20 as blocking agents were satisfactory. The adsorbability of treated polstyrene micr-otiter plates with soap power, detergent, cleaning solution was lower. It was necessary to use new polystyrene microtiter plates in assaying cholera antitoxin.
