流行性出血热病毒结构蛋白的比较研究 被引量:2

摘要 EHFV标准株A9,R22和鲁宁-84刘、鲁宁-84肖、R-36的Vero E6细胞培养液经蔗糖密度梯度离心和免疫沉淀后进行SDS-PAGE,结果均存在NP、G_2和G_1 3种结构蛋白,但不同型别EHFV的结构蛋白分子量略有差异,经Western-blot分析发现5株EHFV的NP至少存在50,49和52kd 3种分子量,同时发现R-36株NP与抗姬鼠型和家鼠型EHFV-NP的McAb均有较强反应,证明属于混合型。 Epidemic hemorrhagic fever virus(EHFV)standard strains A 9,R22 and Shandong local strains Luning-84Liu,Luning-84Xiao,R-36 were purified from infected vero E6 cell cultures by sucrose density gradient centrifugation and immunoprecipitation.SDS-PAGE of these preparations showed three bands indicating respectively the structural proteins NP (neucleccapsid protein),62 and G1(envelop glycoproteins)。It was found that there were some differences between the molecular weight(MW)of these structural proteins of apodemus type and house rat type EHFV.Western-blot analyses of structural proteins of 5 EHFV strains showed that there were 3 kinds of MW of NP,which were 50kd,49kd,52kd。It was interesting to find that the NP of R-36 strain both reacted with apodemus type specific McAb and house rat type specific McAb to EHFV-NP.Therefore,we suggested that R-36 strain was mixed EHFV.
