摘要 研究了IPTG诱导重组噬茵体λgtll/恶性疟原虫溶源化大肠杆菌Y1090,肉眼判断终点、自然释放融合蛋白的方法。并用SDS-PAGE和免疫印染证明了该法的可靠性。研究发现,重组噬菌体的用量与培养时间有关,IPTG的浓度对融合蛋白的制备起重大调节作用,大肠杆菌Y1090可用作重组噬菌体溶源化菌。 This paper presents the method of preparation of fusionprotein by natural releasing procedure.Escherichia coli(E·coli)Y1090 were infected with recombinant phages of lambda gt 11/Plasmodium falciparum,and then were induced by isopropylthiogalactose(IPTG).After incubation at 37℃ for about 4-5h,the fusion protein was released and the end-point was judged by eyes directly.The reliability of the method was verified by SDS-PAGE and Western blot.Theresults showed that the volume of recombinants was correlated withcultural time,the concentration of IPTG was important for the production of fusion protein and E·coli Y1090 might be used as recombinant phage lysogenic bacteria.
