摘要
本研究结果表明:(1) 花蕾长度为1.5~2.0mm、花粉发育时期为单核中期时,愈伤组织诱导率最高;(2) 接种日期影响愈伤组织诱导效果。5月初接种的花药愈伤组织诱导率明显高于10月份;(3) 石刁柏不同品种、倍性及植株间,愈伤组织诱导率有显著差异,UC—72品种的诱导率高于Mary washington 500,二倍体植株诱导率高于四倍体植株;(4) 诱导愈伤组织以1/2MS+BA1.0~2.0mg/l+NAA2.0mg/l+蔗糖4%较好,茎叶分化培养基为MS+BA1.0mg/l+NAA0.2mg/l+蔗糖2%。
Following results are obtained by the anther of asparagus (As paragus officinallis L.): (1) The inducing rate of callus is the highest when the flower bud is 1.5—2.0mm long and the pollen in it is in the mid-period of mono-nucleus development. (2) The inoculating date affects the efficiency on inducing of callus. The callus inducing rate of anther is higher in the beginning of May than in October significantly. (3) There is obvious difference of callus inducing rate between different cultivars, different ploidy and different single plant in the same cultivar. The rate of 'UC-72' is higher than that of 'Mary Washington 500', with the rate of diploid plant higher than that of tetraploid plant. (4) The medium of 1/2MS supplemented with 1.0-2.0mg/1BA, 2.0mg/1NAA and4% sucrose is suited for inducing callus,while that of MS supplemented with 1.0mg/1BA, 0.2mg/INAA and 2% sucrose is applicable for differention of stem and foliage.
