肥胖合并高胰岛素血症患者大网膜脂肪组织中肿瘤坏死因子-α基因的表达及意义 被引量:2

摘要 目的 研究大网膜脂肪组织中肿瘤坏死因子 (TNF) - α基因的表达及其在肥胖和胰岛素抵抗发病机制中的作用。 方法  (1)选择肥胖合并胰岛素抵抗患者 (体重指数 ,BMI<30 kg/m2 ,空腹血浆胰岛素 >2 0 μIU/m l,疾病组 )和对照组 (BMI<2 5 kg/m2 )各 10例 ,抽提大网膜脂肪组织 RNA。(2 )用半定量 RT- PCR方法研究两组大网膜脂肪组织中 TNF- α基因的表达。 结果  (1)疾病组中 TNF- α RNA水平显著高于对照组 (P<0 .0 1)。 结论 (1)疾病组大网膜脂肪组织中 TNF- α表达增多 ,由于 TNF- α能抑制脂肪细胞堆积 ,因此这可能是机体的保护反应 ;(2 )肥胖患者中可能存在 TNF- α限制脂肪堆积作用减弱 。 Objective\ To study the effect of tumor necrosis factor(TNF) α gene in pathogenesis of obesity and insulin resistance.\ Methods\ (1)The initial study comprised 10 obesity and insulin resistance patients(body mass index, BMI≥30 kg/m\+2, fasting blood insulin level >20 μIU/ml) and 10 normal individuals(BMI <25 kg/m\+2), RNA was extracted from omental adipose tissue.\ (2)Semi quantitative RT PCR was conducted to compare the expression of TNF α gene in omental adipose tissue of obesity and insulin resistance patients with normal individuals.\ Results\ In omental adipose tissue, the level of TNF α RNA was higher in obesity and insulin resistance patients than in normal controls.\ Conclusion\ (1)TNF α RNA level increases in omental adipose tissue of obesity and insulin resistance patients.\ (2)In obesity patients, the action of TNF α in limiting fat accumulation may be abnormal and this could result in obesity.
