高效液相色谱法测定五味子乙素兔体内浓度

HPLC Determination of Schisandrin B in Rabbit Plasma

目的 :测定中草药有效成分五味子乙素在兔体内的浓度。方法 :采用反相高效液相色谱法 ,流动相为甲醇 -水 (75∶2 5 ) ,流速 :1mL·min- 1 ,检测波长 :2 5 4nm ,柱温 :32℃。血样预处理采用乙腈去蛋白。结果 :在 0 1～ 4 0 μg·mL- 1 浓度范围内线性关系良好 (r=0 9986 )。平均回收率为 95 33%。日内日间精密度在 10 %以内。结论 :高效液相色谱法可用于测定五味子乙素体内浓度。

Objective:To determine the rabbit plasma concentration of schisandrin B that is active constituent of traditional Chinese medicine Wuling capsule.Methods:A reversed-phase HPLC was used to determine schisandrin B concentrations in rabbit plasma.The mobile phase consisted of methanol-water(75∶25) and the flow rate was 1 mL·min -1.The detective wavelength was at 254 nm,the temperature of column was 32 ℃.Acetonitrile was used to precipitate protein in rabbit plasma.Results:Within 01～40 μg·mL -1,schisandrin B had a good linearity(r= 0998 6).The mean recovery of schisandrin B was 9533%.The within-day precision and day-to-day precision RSD were less than 10% at three levels.Conclusion:The HPLC method could be used to determine concentrations of schisandrin B in rabbit plasma.
