摘要
目的 :用高效液相色谱法测定血浆中鹿蹄草素的浓度。方法 :采用YWG -C1 8色谱分析柱 (4 6mm× 2 5 0mm ,5 μm) ,WatersTM486可变波长紫外检测器 ,甲醇 -水 (4 0∶6 0 )为流动相 ,香草醛为内标。血液样品用乙醚提取纯化后进样 ,在2 2 0nm波长处检测。结果 :鹿蹄草素线性范围为 1 0~ 10 0 0mg·L- 1 (r=0 995 3) ,最低检测量 0 1μg ,最低检出浓度 5 0mg·L- 1 ,平均方法回收率 98 36 % ,精密度考察鹿蹄草素日内、日间RSD均小于 4%。结论 :本方法专属性高 ,快速 ,准确 ,重复性好。
Objective:A HPLC method is described for the analysis of pyroline in pig plasma.Methods:The YWG-C 18 column (46 mm×250 mm,5 μm) was used,The mobile phase consisted of methanol-water (40∶60),at a flow rate of 07 mL·min -1.Vanillin was adopted as the internal standard at UV absorption of 220 nm.Results:The ranges of pyroline calibration curves were 10~1000 mg·L -1(r=0995 3). Detection limits were 0\^1 μg and the lowest detectable concentration of pyroline was 5\^0 mg·L -1,while the average recovery of the method was 98\^36%.The RSD of intra-day and inter-day assays was all less than 4%.Conclusion:The method is quick,precise and good repeat.
