两种灵芝硒多糖分离纯化及性质鉴定

Isolation, purification and property identification of two kinds of polysaccharide containing selenium in Ganoderma lucidum

从灵芝 ( Ganoderma lucidum)加硒深层培养的菌丝 (含 Se 1 60 0μg/ g)中 ,经沸水、碱沸水提取 ,醇析 ,透析 ,Sevage法脱蛋白 ,DEAE-cellulose柱层析纯化后 ,得 Se GLP-1和 Se GLP-2两种灵芝硒多糖 .经 Sephadex G-1 0 0、聚丙烯酰胺凝胶电泳和紫外光谱分析鉴定 ,Se GLP-1和Se GLP-2为均一级分 . GC与 PC分析表明 :Se GLP-1含 5种单糖 Gal、Man、Glc、Xyl、Rha,n( Gal) :n( Xyl) :n( Glc) :n( Rha) :n( Man) =0 .2 3 :0 .72 :1 .0 0 :0 .2 5:0 .0 9. Se GLP-2含有 4种单糖 Gal、Xyl、Glc、Rha,n( Gal) :n( Xyl) :n( Glc) :n( Rha) =2 5.59:1 6.3 0 :1 .0 0 :0 .86.经红外光谱分析 ,确定 Se GLP-1和 Se GLP-2均是由α-糖苷键连接的吡喃多糖 .

Two kinds of polysaccharide containing selenium SeGLP 1 and SeGLP 2 were obtained from fermented mycelia of Ganoderma lucidum rich in selenium. They were isolated and purified by chromatography of DEAE cellulose column. The results of chromatography of SephadexG 100 column, polyacrylamide gel electrophoresis and UV spectra showed that SeGLP 1 and SeGLP 2 were purification profile. Gal, Man, Glu, Xyl and Rha were identified as component sugars of SeGLP 1; Gal, Xyl, Glu and Rha were identified as component sugars of SeGLP 2 by TLC and GC. The quantitative analyses of SeGLP 1 and SeGLP 2 show that the molar ratio of Gal, Xyl, Glu, Rha, Man is 0.23∶0.72∶1.00∶ 0.25∶ 0.09 and Gal, Xyl, Glu, Rha is 25.59∶16.30∶1.00∶0.86. All of them are pyranopolysaccharide by α glycosidic linkage.
