摘要
目的 :在体实验研究神经生长因子 (NGF)对部分视神经损伤的保护效应。方法 :建立视神经不完全损伤模型 ,球后注射神经生长因子 ,记录F VEP ,定量RGCs和髓鞘以评价治疗效果。结果 :与正常组相比 ,NGF组F VEP波形平缓 ,N1和P1潜伏期延长 ,但无显著性差异 ,N1-P1、P1-N1幅值降低、N2 潜伏期延长 ,有显著性差异。NGF对神经节细胞挽救率是19 3%。NGF组未发生溃变的轴突数是损伤未治疗组的 1 83倍。结论 :NGF可挽救视网膜神经节细胞的存活率 ,减少轴突溃变 ,对视网膜神经节细胞继发变性有明显的保护作用。
Objective:To observe in vivo the effects of nerve growth factor(NGF) on the survival of retinal ganglial cell (RGCs) and regeneration of crushed optic nerve axons in adult ratsMethods:After optic nerve crush,NGF was administered by retrobublar injection.The visual evoked potential responses to flash (F-VEP) were recorded,and the numbers of RGCs and axon countedResults:In NGF group,the latent period of F-VEP extended and the swings of F-VEP were depressed,showing no significant difference from normal N 1 and P 1 and distinct difference from normal N 1-P 1、P 1-N 2 and N 2NGF rescue about 193% RGCsNGF prevent axon injury about 183-foldConclusion:The findings of this study demonstrate that treatment with NGF is able to protect neurons and axons from degeneration following optic nerve crush
