摘要 目的:初步了解肺鳞癌及癌旁组织的基因表达谱的改变并探讨cDNA 微阵列表达分析滤膜在肿瘤研究中的应用价值。方法:提取肺鳞癌及癌旁组织总RNA,逆转录成a-32P-dATP,标记的cDNA探针,与含有588个肿瘤相关基因的AtlasTM Human Cancer cDNA Expression Array杂交。放射自显影后利用扫描杂交信号灰度的强弱在专用的 AtlasImage TM软件分析后得出结果。结果:肺鳞癌组织中,肿瘤坏死因子受体(TNFR)等 16个基因表达上调,白介素-13前体等8个基因表达下调。结论:肺鳞癌恶性表型是由其发生过程中多个基因表达的复杂变化决定的。Atlas微阵列表达分析滤膜是研究基因表达谱的较好方法。 Objective To describe the specific expression profile changes in the lung squamous cell carcinoma tissues and their almost normal tissues and explore the value of cDNA microarray in the field of cancer study. Methods: The cDNA probes synthetized from the total RNA that were extracted respectively from the carcinoma and almost normal tissues were hybridized with Atlas human cancer cDNA expression microarray membranes containing 588 genes. The scanned autoradiograms were analysed with Atlas Image software. Results: Sixteen genes including TNFR were found up-regulated in carcinoma while & genes down-regulated including IL-13. Conclusion: The complex alterations of gene expression underlie the development of malignant phenotype of lung squamous cell carcinoma and the Atlas cDNA expression array is a useful method for describing the expression profiles.
