摘要
目的 探讨耐多药结核分枝杆菌耐药基因突变与耐药性的关系。方法 用聚合酶链反应和聚合酶链反应单链构象多态性分析对 12 8株结核分枝杆菌临床分离株耐利福平 (rpoB)、链霉素(rpsL)和异烟肼 (KatG)基因检测。结果 38株敏感株中 ,各有 1株 (2 6 % )测出rpoB、rpsL和KatG突变 ;90株耐多药株中 ,分别有 81株 (90 0 % )、71株 (78 9% )和 6 3株 (70 0 % )测出rpoB、rpsL和KatG突变 ;有 77株 (85 6 % )同时测出 2种以上耐药基因突变 ;10株耐其他药物株中 ,仅有 1株测出KatG基因突变。结论 85 6 %的耐多药结核分枝杆菌存在 2种以上耐药基因突变 。
Objective To investigate the relationship between multi drug resistance and three gene mutations Methods The RFP (rpoB), SM (rpsL) and INH (KatG) genes in 128 M tuberculosis isolates were analyzed with PCR and PCR SSCP Results The mutation in rpoB、rpsL and KatG gene was respectively detected in 1 of 38 drug susceptible isolates, in 81 (90 0%)、 71(78 9%) and 63 (70 0%) of 90 multi drug resistance In 90 multiple resistant strains, 77 (85 6%) showed mutations in more than two genes associated with resistance to each of these three drugs One of 10 nono resistant isolates not to RFP SM and INH drug revealed only mutation in KatG gene Conclusion 85 6% of multi drug resistance in M Tuberculosis showed mutations in more than two drug resistant genes, which are associated with concentration of drug resistance
