HPLC法同时测定复方头孢氨苄胶囊中头孢氨苄和甲氧苄啶的含量

Simultaneous Determination of Cefalexin and TMP in Compound Cefalexin Capsules by HPLC

目的 :以HPLC法同时测定复方头孢氨苄中的头孢氨苄与甲氧苄啶的含量。方法 :HPLC外标法 ,采用岛津C18反相色谱柱 ,以V(水 )∶V(甲醇 )∶V(3 .86 %乙酸钠溶液 )∶V(4%乙酸溶液 ) =742∶2 40∶15∶3为流动相 ,流速 1.2mL/min ,在波长 2 5 4nm处进行检测。结果 :本方法可同时测定两组分的含量。被测组分能很好地分离 ;头孢氨苄在 37.19～ 371.9μg/ml,甲氧苄啶在 8.0～80 .0 μg/ml范围内 ,峰面积与其浓度呈良好线性关系 ;平均回收率依次为 99.0 2 % ,RSD =0 .19% ;99.81% ,RSD =0 .15 %。结论 ,以HPLC外标法同时测定复方头孢氨苄中两组分的含量 ,方法简便、可靠 ,重现性良好。

Objective:To determine the content of cefalexin and trimethoprim (TMP) in compound cefalexin capsules,simultaneously by HPLC. METHOD: On the basis of external standard method ,the C 18 reversed phase column was used. Water methanol 3.86% sodium acetate solution 4% dilute glacial acid (742∶240∶15∶3) was used as mobile phase, with the flow rate of 1.2 mL/min. The detection was carried out with an UV detector at λ 254 nm. Results: The separation and determination of the two constituents could be accomplished simultaneously. The calibration curves were linear in the range of 37.19～371.9 μg/mL for cefalexin and 8.0～80.0 μg/mL for TMP. The average recoveries of cefalexin and TMP were 99.02%( RSD=0.19%) and 99.81%(RSD =0.15%) respectively. Conclusion: This method was simple,rapid and accurate for simultaneous determination of the two constituents in compound cefalexin capsules.
