摘要
目的 探讨镁 (Mg2 + )对缺氧缺血性脑损伤的神经保护作用及其与神经细胞凋亡的关系。方法 结扎新生 7日龄Wistar大鼠左颈总动脉后吸入 8%浓度氧 2h制成标准化动物模型 ,72h后断头取脑。用原位末端标记技术和苏木素 伊红染色分别对对照组、缺氧缺血组和硫酸镁治疗组动物脑组织中凋亡细胞数量和脑组织神经病理改变进行观察比较。结果 缺氧缺血后脑组织内末端标记阳性的凋亡细胞明显增加 (t =- 5 6 6 1,P <0 0 0 1) ,神经细胞变性显著。硫酸镁治疗组标记阳性的细胞数比同时间的缺氧缺血组显著降低 (t =10 36 ,P <0 0 0 1) ,神经细胞变性坏死减少。结论 镁对新生鼠缺氧缺血性脑损伤有保护作用 ,其作用与抑制细胞凋亡有关。
Objective To investigate the neuroprotective effect of magnesium sulfate on hypoxic-ischemic brain damage and its relation with apoptosis.Methods The HIBD model was produced using 7-day old Wistar rats subjected to unilateral common carotid artery ligation followed by hypoxic state(8% oxygen+92% nitrogen at 37℃) for 2 hours.The rats were divived into three groups:control group、HI group and MgSO 4-treated group.The rats were sacrificed 72 hours after hypoxic ischemia.Hematoxylin-Eosin(HE) staining and terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end -labeling (TUNEL) were used to evaluate the histopathological features and the number of apoptic cells in brain tissue of the three groups.Results It was found that the numbers of apoptic cell were markedly increased in hypoxic-ischemic brain tissue(t=-5661,P<0001) and decreased significantly in MgSO 4-treated group compared with that in HI group(t=1039,P<0001).The neuronal degenerations in cortex were reduced in MgSO 4-treated group.Conclusion The data indicate that magnesium has protective effect on HIBD,which has relation with inhibiting apoptosis.
