摘要
目的 :探讨细胞凋亡与博莱霉素诱导大鼠早期肺纤维化及bax ,bcl 2对博莱霉素大鼠肺纤维化细胞凋亡的作用 .方法 :通过复制博莱霉素大鼠肺纤维化模型 ,采用TUNEL、免疫组织化学、原位杂交及透射电镜等技术 ,观察博莱霉素大鼠肺组织细胞凋亡的变化及bax ,bcl 2 ,bcl 2mRNA和蛋白的表达 .结果 :博莱霉素诱导的大鼠肺纤维化细胞凋亡明显增加 ,肺上皮细胞bax蛋白表达明显上调 ,bcl 2mRNA及蛋白表达明显减弱 ,间质细胞bcl 2mRNA及蛋白表达增加 .结论 :肺组织细胞凋亡及bcl 2表达下调可能参与博莱霉素诱导大鼠肺纤维化的发生 ,凋亡相关基因bax ,bcl
To study the morphology,the incedence of apoptosis and the expression of bax,bcl 2 and bcl 2mRNA in bleomycin induced lung injury in rats,wistar rats were intratracheally instilled with bleomycin(5?mg/kg of body weight).The animals injected with sterile saline were anesthetized and killed at 14 days after bleomycin instillation.HE staining,electron microscopy,terminal deoxynucleotigyl transferase mediated dUTP nick labeling(TUNEL),immunohistochemisty(IHC)and in situ hybridization(ISH)methods were used.The results show that a single instillation of bleomycin leads to the appearance of apoptosis in bronchial and alveolar epithelia cells.The expression of bcl 2 and bcl 2mRNA were downregulated and the expression of bax was upregulated in the alveolar epithelia cells.Those proves that the bcl 2/bax system may play a role in the apoptosis of bronchial and alveolar epithelia cells in this disorder.
