摘要
本文首先报道培养的新西兰大白兔的血管平滑肌细胞(SMC)能对WST-1进行还原代谢,并联合应用WST-1和BrdU ELISA测定同一标本的SMC的增殖及其代谢活性。结果表明:随着胎牛血清浓度的增加,SMC活细胞数目、WST-1和BrdU的OD值逐渐增多。说明WST-1和同一标本的WST-1和BrdU ELISA联合测定方法是检测SMC增殖的指标,能客观地反映SMC的DNA合成和代谢活性。
WST-1 that could be metabolized by the cultured smooth muscle cells( VSMC)of New Zealand white rabbit was firstly reported in this paper. In the same sample, WST-1 and BrdU colorimetric ELISA were also used simultaneously to measure the DNA synthesis and metabolic activity of VSMC. The results showed the viable cell number, the OD of WST-1 and BrdU were increased respectively with the increasing concentration of fetal calf serum in VSMC. It demonstrates that WST-1. WST-1 and BrdU ELISA simultaneous measurement can reflect objectively the DNA synthesis and metabolic activity of VSMC and detect proliferation of the VSMC.
