摘要
通过对敲除磷脂酶 C-γ1 基因的鼠成纤维细胞株及其转染磷脂酶 C-γ2 基因后的迁移指标的统计学分析 ,揭示出磷脂酶 C-γ1 和γ2 在 PDGF引起的细胞迁移信号传导中均有正调控作用 ,前者作用显著而后者作用微小 ,并且仅在磷脂酶 C-γ1 功能缺失的条件下方可发挥出来 ;同时进一步证明敲除磷脂酶 C-γ1 基因的细胞株中 ,未见出现磷脂酶 C-γ2 的代偿 ,说明磷脂酶 C-γ2 与 γ1 在功能上不可相互取代 ,反映了一种信号可由多种途径传导即细胞信号传导的交互性。
By statistical analysis of the migration indices of phospholipase C γ 1 gene knocked out and phospholipase C γ 2 gene vector transfected fibroblasts, we found that phospholipase C γ 1 and phospholipase C γ 2 both shew a PDGF induced positive regulator effect on the migration signal of fibroblast. The effect of the former was significantly stronger than the later and phospholipase C γ 2 played an active role only under the condition with functional deficiency of phospholipase C γ 1. This study also showed no phospholipase C γ 2 compensation in phospholipase C γ 1 gene knocked out cell line, which suggested that these enzymes could bring with independent functions which implied complexity and redundancy in such multiple signal transduction pathway.
