摘要
^(199)Au具有很好的核性质,适合于标记单克隆抗体,用于肿瘤的放免诊断与治疗。为了寻求稳定地用金标记单克隆抗体的方法,本文研究了采用还原剂NaBH_4或NaHSO_3还原蛋白质上的二硫键为游离的巯基,再与Au(Ⅰ)络合的标记方法。用此种方法成功地制备了^(199)Au标记的马拉人IgG多克隆抗体,并保持了抗体的免疫反应的活性。
198Au has good nuclear property for application to the labelling of monoclonal antibody, eventually to the diagnosis and the therapy of tumours. An effective method is found to label antibody with Au by using reducing agents such as NaBH4 ,NaHSO3 to reduce disulfide in the protein into free thiols with which Au(Ⅰ) could be bound. By this method 199Au-labelled horse polyclonal antibody against human IgG without loss of its immunoreactivity is prepared successfully. This method could also be employed to label monoclonal antibody.
