摘要
研究了^(125)IUdR常用示踪量对肿瘤细胞生物学性能的影响。结果显示:示踪量为0.1934=dpm/cell-DNA时,微核率已显著增加;示踪量为1.3323dpm/cell-DNA时,体外生长曲线的斜率明显下降,T_D值显著增加;示踪量为1.1795 dpm/cell-DNA时,肺转移结节数显著减少;示踪量分别为0.1934和0.3079dpm/cell-DNA时,对皮下局部生长已有抑制作用。提示在示踪试验中应尽量控制^(125)IUdR的示踪量,最好能控制在0.2dpm/cell-DNA以下。
The effect of tracer doses of 125IUdR on the biological properties of tumor cells was studied.Experimental results demonstrated that the micronucleus rates of B16 melanoma cells increased significantly when the dose of 125IUdR was 0.1934 dpm/cell-DNA. The doubling time of cells obviously increased and the regression coefficient of in vitro cell growth curve decreased significantly at 1.3323 dpm/cell-DNA. Lung metastatic colonies of B16 melanoma decreased significantly when the dose of 125IUdR was 1.1795 dpm/cell-DNA. The subcutaneous growth of tumor cells have been inhibited obviously when the dose was 0.1934 or 0.3079 dpm/cell-DNA. Therefore, the dose of 125IUdR in tracer experiments should be carefully controlled, preferably below 0.2 dpm/cell-DNA.
