摘要
目的探讨同种异体软骨细胞-聚羟基乙酸 (PGA)复合物在有免疫力的动物体内对喉甲状软骨的修复能力。方法消化分离乳兔肋软骨,获取种子细胞,体外培养,收养第 2~ 3代培养细胞,接种于经多聚赖氨酸处理后的 PGA三维可降解生物支架料上。结果复合物体外培养 7~ 10d,可见呈蜘蛛网状基质产生。术后 4周,修复区愈合良好,无瘢痕及坏死现象;组织学观察可见愈合面有软骨细胞生成和基质分泌。结论软骨细胞- PGA复合物可用于喉软骨缺损的修复,修复区愈合良好,无明显免疫排斥反应。
Objective To explore the ability to repair laryngeal cartilage defects using chondrocyte- polyglycolic acid(PGA)complexes.Method Chondrocytes were isolated from costa- cartilage of infant rabbits byⅡ type collagenase digestion.they were cultured in vitro for 2~ 3 generations with Ham- F12,and then the cells were cultured in PGA scaffolds.Result cartilaginors matrix was observed 7~ 10 days after culturing.4weeks after operation,laryngeal cartilage defects were initally filled with new cartilage.No scar and necrosis in operative areas were found.More chondrocytes and matrix were observed in reparative areas.Conclusion Chondrocyte- PGA complex can be used as reparative material for laryngeal thyroid cartilage defect.
