摘要
目的 :通过测定SD大鼠坐骨神经离断后比目鱼肌 (SOL)和趾长伸肌 (EDL)肌纤维横截面积 ,观察持续给睫状神经营养因子 (CNTF)对去神经骨骼肌萎缩的影响。结果 :SD大鼠坐骨神经离断后 ,持续给 0 .2mg/kg.d的CNTF 2 0天后 ,损伤侧SOL和EDL肌纤维横截面积分别比实验对照组高 2 7% (P <0 .0 1 )和 1 4 % ,SOL肌纤维横截面积比EDL增加的幅度大 ,而给予 0 .0 5mg/kg .d的CNTF 2 0天后 ,动物肌纤维横截面积与实验对照组无明显差异。结论 :CNTF可显著改善坐骨神经离断后SD大鼠骨骼肌的萎缩 ,并且CNTF效应的强弱与用药剂量和肌肉类型有关 ,0 .2mg/kg .dCNTF作用明显强于 0 .0 5mg/kg .dCNTF ,慢肌 (SOL)比快肌 (EDL)
The effect of ciliary neurotrophic factor (CNTF) on the denervetde skeletal muscle atrophy was studied by detecting fiber cross-sectional areas soleus muscle (SOL) and extensor digitorum longus (EDL) in nerve transected SD rats.SD rats with sciatic nerve transected were administrated CNTF (0.2mg/kg)continuously for 20 days.It were found that in the fiber cross-sectional areas in injury side,SOL and EDL raised by 27% ( P <0.01) and 14%,with a bigger increase in SOL fiber cross-sectional areas than EDL compared with that of model group.But no obvious changes was observed in animal administrated with CNTF of 0.05 mg/kg.CNTF attenuated skeletal muscle atrophy significantly in sciatic nerve transected SD rats,so it could be said that CNTF had atrophic effect on denervated skeletal muscle.The effect may be in a dosage dependent manner.And it was related with the type of muscle,because the myotrophic effect of 0.2 mg/kg CNTF was more effective than that of 0.05 mg/kg CNTF,and slow twitch muscle (SOL) reacts to CNTF stronger than quick twitch muscle did (EDL).
