摘要
目的 观察静态负荷对家兔骨骼肌线粒体功能的影响 ,探讨肌肉损伤的机理。方法 以原子吸收光谱法测定家兔腰肌线粒体的Ca2 +含量 ;根据酶促水解ATP生成磷的含量表示Ca2 + Mg2 + ATP酶活性 ;用硫代巴比妥酸 (TBA)反应法测定丙二醛 (MDA)含量。结果 施加静态负荷 1、4周时家兔腰肌线粒体的Ca2 +含量显著高于对照组 (P <0 0 1) ;线粒体Ca2 + Mg2 + ATP酶活性在施加静态负荷 1周时明显降低 (P <0 0 1) ;施加静态负荷 1、4周时线粒体MDA含量显著高于对照组 (P <0 0 5 )。结论 静态负荷致肌肉损伤过程中线粒体功能下降 。
Objective To study the mechanism of muscle injury by observing the effects of static load on mitochondria function in rabbit muscle. Methods Ca 2+ concentration of mitochondria in rabbit muscle was measured by atomic absorption spectrophotometry. The contents of phosphorus from ATP were measured by the activity of Ca 2+ Mg 2+ ATPase as the indicator of zymohydrolysis. The malondialdehyde (MDA) contents of mitochondria were measured by the method of thiobarbituric acid reaction (TBA). Results The Ca 2+ concentrations of mitochondria were significantly higher than those of control groups respectively after 1 week and 4 weeks ( P<0 01 ). After 1 week the Ca 2+ Mg 2+ ATPase activity of mitochondria was lower than that of control group ( P<0.01 ). The contents of MDA were significantly higher than those of controls after 1 week and 4 weeks ( P<0.05 ). Conclusion The muscle injury might be related to the decrease of the function of mitochondria.
