单克隆抗体亲和层析最适洗脱液筛选研究

摘要 本文介绍一种筛选单克隆抗体(McAb)亲和层析最适洗脱液的方法,一种经过改进的双抗体夹心酶联免疫吸附检测法(DAS—ELISA),其中以联有一种McAb的经CNBr活化的Sepharose凝胶为固相,另一种McAb标上辣根过氧化物酶作为标记抗体。我们在以单抗亲和层析纯化促卵泡激素(FSH)的工作中使用该方法进行了最适洗脱液的筛选,取得了很好的结果。该方法简便、快速、可靠。 An improved method of screening the optimal eluent in monoclonal antibody (McAb) affinity chromatography is developed and recommended. It is a double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), in which the solid phase is a CNBr-activated Sepharose 4B gel linked with a McAb and another McAb labeled with horseradish peroxidase (HRP)is used as the marker antibody. The method has been applied to screen an optimal eluent in an affinity chromatography of purifying a kind of follicle-stimulating hormone (FSH)and obtained excellent results. The method is simple, rapid and reliable.
